your main screen will advise your CSRs/Dispatchers that the date selected is not in the preferred days but it will let you book it, if you go to scheduler assistant it will let you book the job and still give you the best route, so the answer is no, scheduler assistant doesn't take it into account. Service days is only a "warning" to your CSRs/Dispatchers just like skills
